Rapid and Direct Compositional Analysis of Poly(3-hydroxybutyrate- co -3-hydroxyvalerate) in Whole Bacterial Cells by Thermally Assisted Hydrolysis and Methylation–Gas Chromatography

摘要: Thermally assisted hydrolysis and methylation-gas chromatography (THM-GC) in the presence of an organic alkali was applied to direct analysis copolymer composition for poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] accumulated whole bacterial cells. Cupriavidus necator grown on a liquid medium with different molar ratios valeric acid (V) glucose (G) order control compositions P(3HB-co-3HV) produced Trace amounts (0.03 mg) dried cells were directly subjected THM-GC tetramethylammonium hydroxide (TMAH) at 400°C. The obtained chromatograms clearly showed nine characteristic peaks, attributed THM products from 3HB 3HV units polymer chains, without any appreciable interference by matrix components. Based these peak intensities, determined rapidly using cumbersome sample pretreatment. Moreover, thus good agreement those conventional technique.
