摘要: Cloned human CD4+ T cell lines specific for the house dust mite Dermatophagoides pteronyssinus were used to map minimal activation-inducing epitopes on Group I allergen in D. extracts (Der p I) molecule. Most of these Der I-specific clones expressed different TCR V alpha and beta gene products. Using recombinant deletion proteins, three identified molecule; p45-67 p117-143 recognized by HLA-DR7-restricted cells, whereas p94-104 was context HLA-DR2, DRw11 (DR5), -DR8 molecules. This degenerate class II MHC restriction appears be due shared Phe Asp residues at positions 67 70, respectively, third variable domain HLA-DR chain. All induced Th2-like cytokine production profiles clones, which characterized very high levels IL-4 IL-5, as compared with those secreted tetanus toxin-specific derived from same patients, but no or low amounts IL-2 IFN-gamma. profile was, however, not an intrinsic property dependent upon their mode activation. Stimulation Con A also measurable IFN-gamma, activation TPA calcium ionophore A23187 resulted IL-4, IL-2, These results indicate that are defective capacity produce Th1 cytokines.
