Characterization and transcriptional analyses of cDNAs encoding three trypsin‐ and chymotrypsin‐like proteinases in Cry1Ab‐susceptible and Cry1Ab‐resistant strains of sugarcane borer, Diatraea saccharalis

摘要: Diatraea saccharalis is a major corn borer pest. Midgut serine proteinases are essential for insect growth and development. Alteration of midgut responsible Bt resistance development in some species. To clone trypsin chymotrypsin cDNAs to test if the Cry1Ab D. associated with changes proteinases, total tryptic chymotryptic activities, cDNA sequences, gene expressions three genes were comparatively examined between Cry1Ab-susceptible (Cry1Ab-SS) Cry1Ab-resistant (Cry1Ab-RR) strains. Full-length encoding trypsin- chymotrypsin-like sequenced from Cry1Ab-SS Cry1Ab-RR larvae. These code active forms all functional motifs, including signal peptide, conserved His-Asp-Ser catalytic triad, pairs cysteines disulfide bridge configurations, substrate specificity determination residues. In general, putative protein sequences highly similar strains, except few nucleotide predicted amino acid substitutions, whose function need be further clarified. Total activities also Transcriptional levels had numerical difference but was not statistically significant. Data suggest that significantly these trypsins chymotrypsins. Results clarified role six provided foundation continuing examination potential involvement other important biochemical processes.
