Polydatin: a new therapeutic agent against multiorgan dysfunction.

摘要: Abstract Background Polydatin (PD), a monocrystalline and polyphenolic drug isolated from traditional Chinese herb (Polygonum cuspidatum), is protective against mitochondrial dysfunction has been approved for clinical trials in the treatment of shock. However, whether administration PD therapeutic effect on multiple-organ syndrome (MODS) requires investigation. Material methods MODS was induced Sprague–Dawley rats via hemorrhage ligation puncture cecum–induced sepsis. The were divided into three groups as follows: MODS + PD, MODS + normal saline, control group (no treatment). Survival time, blood biochemical indexes, histopathologic changes various organs evaluated; serum oxidative stress (advanced protein products [AOPPs]) proinflammatory cytokines (tumor necrosis factor-α, interleukin 1β, 6) assayed using enzyme-linked immunosorbent assay. Apoptosis-related expression (B-cell lymphoma-2 [Bcl-2] Bax) by immunohistochemical Western blotting methods, whereas caspase-3 activity spectrophotometry. Results improved organ function, prolonged survival reduced incidence levels AOPPs cytokines. It also decreased Bax increased Bcl-2 kidney liver. Conclusions may serve potential MODS, it suppresses stress, inhibits inflammatory response, attenuates apoptosis, protects dysfunction.
