Skin differences based on age and chronicity of ultraviolet exposure: results from a gene expression profiling study

摘要: Summary Background  Skin ageing represents an inevitable physiological consequence of getting older but the impact on personal health and wellbeing can be significant, therefore considerable efforts have been made to understand biology pathophysiology skin try identify new targets that might offer therapeutic intervention prevention. Objectives  This study was designed differences at gene expression level between young old, sun-exposed sun-protected skin. Methods  We generated transcriptomic data from old sites (10 samples each) using HG-U133 Plus 2.0 Affymetrix GeneChips. The were analysed hierarchical clustering, theme analysis interaction mapping regulated pathways, processes potential for therapy. Results  With 54 613 probe sets GeneChip, 2731 significant would expected by chance (at P = 0·05), we noted 13 640 significantly different comparing arm vs. (photoageing), 7215 buttock comparison (intrinsic ageing). In both types there reduced many genes implicated in lipid biosynthesis epidermal differentiation with functional relevance barrier integrity maintenance. Increased contributing oxidative stress decreased antioxidant defences also common ageing. Differences intrinsic photoageing mainly extracellular matrix interstitial collagen increased elastic tissue photoageing. Conclusions  Collectively, identified biomarkers aged skin, particularly involving abnormalities proteases, proteins inflammation. These findings prospect more specific development based improved understanding
