摘要: Background An altered Kupffer cell (KC) response is thought to be responsible for the characteristic phenotype observed after biliary obstruction: a marked by defect in hepatic reticuloendothelial system and hypersensitivity endotoxin. Few studies, however, have directly examined KC function. We sought define specific alterations function that occur obstruction. Methods KCs were isolated from female C57BL/6 mice 4 days sham or common bile duct ligation (CBDL) operation. Phagocytosis, oxidative burst potential, intracellular bacterial killing measured as markers of The endotoxin was assessed measuring tumor necrosis factor alpha interleukin 6 levels media stimulation with lipopolysaccharide (LPS) LPS plus LPS-binding protein (LBP). Results CBDL demonstrated significant increase phagocytic ability significantly decreased baseline stress, compared Shams. equivalent higher KCs. also increased numbers viable bacteria infection; it unclear if this finding represents impaired phagocytosis bacteria. With respect endotoxin, found less sensitive stimulatory effects alone but exquisitely LBP. LBP elevated animals, dose-dependent, exaggerated administered Conclusions clearly Phagocytic actually increased, although kill within phagosome remains ill defined. are LBP, may proinflammatory challenge animals
