摘要: Background: The stability and stoichiometric changes of analytes in plasma serum after prolonged contact with blood cells uncentrifuged Vacutainer® tubes were studied. Methods: We simultaneously investigated the 24 ( a ) b immediate separation (centrifuged twice at 2000 g for 5 min). verified biochemical mechanisms observed analyte change by concomitant measurement pH, P co2, o2. Hemolysis was qualitatively semiquantitatively assessed. All specimens maintained room temperature (25 °C) analyzed duplicate 0.5, 4, 8, 16, 24, 32, 40, 48, 56 h collection. Statistically significant from 0.5 mean determined using repeated-measures ANOVA. limit applied to determine clinically measured analytes. Results: Fifteen showed relevant changes, degree more pronounced most specimens. immediately separated collection stable. Conclusion: Storage beyond caused because glucose depletion Na+,K+-ATPase pump failure; movement water into cells, causing hemoconcentration; c leakage intracellular constituents metabolites. Immediate or provides optimal temperature. When is unavoidable, use recommended higher instability analytes.
