Involvement of phosphatidylinositol 4,5-bisphosphate binding proteins in the generation of contractile oscillations in the Physarum polycephalum plasmodium

摘要: Using the Physarum polycephalum plasmodium, a giant amoeboid cell with strongly pronounced auto-oscillatory mode of motility, which exhibits regularities motile behavior common those tissue cells and has same signal systems, possibility participation phosphatidylinositol 4,5-bisphosphate in regulation contractile activity been studied. The effect neomycin as substrate inhibitor phospholipase C, binds high affinity to membrane, on force oscillations generated by plasmodial strands under isometric conditions after addition protein kinase C inhibitors staurosporine, UCN-01, Ro-318220, separately combination calmodulin calmidazolium, examined. It shown that at pH 7.0 concentrations 0.1–5.0 mM stops for 10–30 min but then they begin gradually resume; oscillation period initial stage restoration is shorter than it was earlier, increases because elongation contraction phase. Analysis data obtained favor assumption membrane contains MARCKS-like proteins C-controlled pools phosphatidylinositol-4,5-bisphosphate, can participate generation auto-oscillations observed plasmodium.
