摘要: Accurate prediction of regional lymph node metastasis (LNM) in endoscopically resected T1-stage colorectal cancers (CRCs) can reduce unnecessary surgeries. To identify miRNA markers that predict LNM CRCs, the study was conducted two phases; (I) classifier construction by miRNA-array and quantitative reverse transcription PCR (qRT-PCR) using 36 CRC samples; (II) validation an independent set 20 samples. The expression potential downstream target genes miRNAs assessed immunohistochemistry. In discovery analysis microarray, 66 were significantly different between LNM-positive negative CRCs. After qRT-PCR validation, 11 consistently significant combined set. Among them, miR-342-3p most one (P=4.3×10-4). Through logistic regression analysis, we developed a three-miRNA (miR-342-3p, miR-361-3p, miR-3621) for predicting yielding area under curve 0.947 (94% sensitivity, 85% specificity 89% accuracy). discriminative ability this system reliable (83% 64% 70% Of targets three-miRNAs, expressions E2F1, RAP2B, AKT1 associated with LNM. conclusion, more accurately than conventional pathologic criteria our results may be helpful to avoid bowel surgery after endoscopic resection early CRC.
