Millicurrent stimulation of human articular chondrocytes cultivated in a collagen type-I gel and of human osteochondral explants.

摘要: Here we investigate the effect of millicurrent treatment on human chondrocytes cultivated in a collagen gel matrix and osteochondral explants. Human from osteoarthritic knee joints were enzymatically released transferred into type-I gel. Osteochondral explants cell-seeded samples in-vitro for three weeks. Samples verum groups stimulated every two days by (3 mA, sinusoidal signal 312 Hz amplitude modulated super-imposed signals 0.28 Hz), while control remained unaffected. After recovery, type-I, type-II, aggrecan, interleukin-1β, IL-6, TNFα MMP13 examined immunohistochemistry real time PCR. With regard to immunostainings 3 D did not show any differences between group. The expression all investigated genes was elevated following treatment. While explant gene col-I, col-II Il-1β nearly unaffected, aggrecan elevated. Following treatment, TNFα, decreased. In general, standard deviations data high, resulting rarely significant results. We conclude that stimulation directly influences cell metabolism.
