Impact of amorphous and semicrystalline polymers on the dissolution and crystallization inhibition of pioglitazone solid dispersions

摘要: Abstract Solid dispersions are used as a useful approach to improve the dissolution rate and bioavailability of poorly water-soluble active pharmaceutical ingredients (APIs). The present study investigated impact amorphous polymers (polyvinylpolypyrrolidone K30 polyvinylpolypyrrolidone K90) semicrystalline (polyethyleneglycol 6000 polyethylene–polypropylene glycol 188) on profile pioglitazone solid crystallization behavior from supersaturable state. Generated by these resulted in significant enhancements compared with crystalline API, whereas differences prepared were not notable. Amorphous themselves superior for enhancement API medium containing pre-dissolved polymers. For maintaining supersaturated state, all observed reduce crystal growth rates drug alone. more effective at inhibiting rates, while much less effective. intermolecular interaction between was enhanced analysis DSC, PXRD, FT-IR that (PVP) most suitable design considering both profiles inhibition effects.
