摘要: A specific and sensitive assay for quantifying the immunosuppressant FK506 its metabolites in blood urine was developed. 32-O-Acetyl synthesized used as internal standard. were purified from samples by solid-liquid extraction injected into a high-performance liquid chromatographic (HPLC) system linked to mass spectrometer (MS) particle-beam interface. The derivatives separated interfering material use of 100 x 4 mm C8 analytical column water/acetonitrile or water/methanol gradient elution; they detected negative chemical ionization with methane reagent gas. limit detection 25 pg standard solution, quantification 250 (extracted 1 mL blood). CV 11.3% at 5 ng, no interferences other drugs found.
