摘要: Video-connected fluorescence microscopy was introduced to study the yeast nuclear chromatin region. It defined as area where a DNA-binding fluorescent probe 4′,6-diamidino-2-phenylindole specifically bound and fluoresced. The 3-dimensional feature of mitotic region deduced by analysing successive video images cell viewed at different angles. By investigating synchronous culture wild-type fission Schizosaccharomyces pombe, we found sequential structural alterations in during mitosis. steps include compaction from regular hemispherical form, formation U-shaped intermediate rapid segregation into 2 daughter forms. Six cs cdc mutants, apparently blocked mitosis, were observed microscopy. Under restrictive conditions their regions exhibited either hemispherical, compact, disk-like, or partially segregated regions. Two mutants showed anomalous locations. results temperature shift-up experiments highly reversible KM52 KM108 strains supported above scheme
