Isolation and structural characterization of human lymphocyte and neutrophil gangliosides.

摘要: Abstract Gangliosides were isolated from purified preparations of human peripheral blood lymphocytes and neutrophils. Structural analyses comparisons performed by direct probe mass spectrometry degradation studies with the following enzymes: Escherichia freundii endo-beta-galactosidase; Clostridium perfringens Arthrobacter ureafaciens neuraminidase; jack bean beta-N-acetylhexosaminidase beta-galactosidase. This combination techniques allowed us to obtain carbohydrate composition sequence information without aid methylation or compositional using only 1-2 mg gangliosides. On basis these we propose that neutrophils have gangliosides structures. NeuAc alpha 2 leads 3Gal beta 1 4Glc 1Cer Structure A ? GlcNAc B Gal 3,4GlcNAc C All three compounds both cell types structure being major lymphocyte ganglioside neutrophil ganglioside. is a novel may represent leukocyte-specific glycosphingolipid. Neuraminidase demonstrated one species each type contains an internally linked sialic acid residue, on thin layer chromatographic analysis this component same as brain ganglioside, GM1 (II3-N-acetylneuraminosyl-gangliotetraosylceramide). In addition, large general ?(Gal 3)n isolated. These results are discussed they relate group antigens specific surface markers in leukocytes.
