摘要: The high sensitivity of PCR compared to the difficulties fecal culture in sheep prompted development protocols for detection Mycobacterium avium subsp. paratuberculosis DNA feces. Although itself is well developed, and does not pose large technical problems, concentrating bacteria from samples that may contain low numbers bacilli using practical methods still main difficulty use this technique. In study, we describe an extraction protocol concentration purification M. compare it with other methods. diagnostic performance freeze-boiling method was evaluated a reference [Vet. Rec. 134 (1994) 95] on group selected different flocks known individual serological, pathological, cultural status. Using, as reference, combination results those conventional methods, showed 94.1%, specificity 92.3%.
