Methylation of the γ-Catenin Gene Is Associated With Poor Prognosis of Renal Cell Carcinoma

摘要: Purpose: γ-Catenin is a cell adhesion protein, and its functional loss associated with tumor invasion metastasis. We hypothesize that (1) promoter CpG methylation regulates the expression function of γ -catenin gene in renal carcinoma (RCC) (2) poor prognosis RCC. To test these hypotheses, we analyzed status correlation clinical outcome Experimental Design: Genomic DNA total RNA were extracted from three cancer lines (A498, Caki-1, Caki-2) 54 RCC tissue samples their corresponding normal kidney samples. Expression was by reverse transcription-PCR immunostaining. Promoter two different methylation-specific PCR (MSP-A MSP-B), results verified sequencing. Results: The demethylating agent (5-aza-2′-deoxycytidine) increased levels mRNA transcript lines. protein significantly reduced compared samples, respectively ( P = 0.029 0.0071, respectively). Because conventional nuclear grade and/or staging system have some limitations to predict precise outcome, first report demonstrating can be an independent superior predictor for survival disease progression.
