Vincetoxicum arnottianum modulates motility features and metastatic marker expression in pediatric rhabdomyosarcoma by stabilizing the actin cytoskeleton.

摘要: Background Prevention of metastatic invasion is one the main challenges in treatment alveolar rhabdomyosarcoma. Still therapeutic options are limited. Therefore, an anti-tumor screening was initiated focusing on anti-metastatic and anti-invasion properties selected medicinal plant extracts phytoestrogens, already known to be effective prevention different cancer entities. Methods Treatment effects were first evaluated by cell viability, migration, invasion, colony forming assays rhabdomyosarcoma line RH-30 comparison with healthy primary cells. Results Initial screenings all substances analyzed this study, identified extract Vincetoxicum arnottianum (VSM) as most promising candidate, harboring highest potential. Those significant anti-motility proven a reduced ability for migration (60%), (99%) formation (61%) under 48 h exposure 25 μg/ml VSM. The restricted motility features due induction stabilization cytoskeleton - actin fibers 2.5-fold longer spanning entire cell. Decreased proliferation (PCNA, AMT, GCSH) altered metastasis (e. g. SGPL1, CXCR4, stathmin) marker expression transcript protein level confirmed lowered tumorigenicity VSM treatment. Finally, alterations metabolism detected metabolites, levels uracil, N-acetyl serine propanoyl phosphate greatest alterations. Compared conventional therapy cisplatin, treated cells demonstrated similar metabolic shutdown metabolism. Primary control not affected Conclusions This study revealed root potential, new migrastatic drug candidate putative pediatric filament stabilizing accompanied marginal effect vitality
