Less is More—Optimization of DNA Extraction from Canine Feces

摘要: Although most DNA crime laboratories may not encounter fecal samples often, they are a familiar sample type in non-human forensic due to their prevalence the environment. Fecal matter can be challenging low numbers of nucleated cells and the presence inhibitors that impede amplification success. Sampling location (internal vs. external), sampling quantity (10–200 mg), various extraction protocols (silica matrix, bead beating, clean-up column) were evaluated maximize yield. The greatest yield intact was obtained using modified silica matrix protocol (VGL-Fecal) on 30–50 mg collected from external surface stool had been dried for 24 h. This optimized applied pilot study where genotyping success evaluated. By optimizing our collection, sampling, procedures, reliabl e method maximizing canine developed.
