Alterations in mRNA expression and protein products following spinal cord injury in humans

摘要: We examined the effects of spinal cord injury (SCI) on alterations in gene expression and respective protein products human skeletal muscle 2 days 5 post-SCI. Biopsies were taken from 9 men 1 woman (n = 10) (43.9 +/- 6.7 years) post-SCI healthy young who served as controls (20.4 0.5 years). Global changes analysed using Affymetrix GeneChips a subsample subjects 3). Candidate genes then pursued via qRT-PCR. Western blotting (WB) was used to quantify candidate genes. Immunohistochemistry (IHC) localize proteins. Groups transcripts showing greatest percentage altered expression, most robust fold-changes, indicative involvement an entire pathway GeneChip included involved ubiquitin proteasome (UPP), metallothionein function, protease inhibition. qRT-PCR analysis confirmed increases for UPP components (UBE3C, Atrogin-1, MURF1, PSMD11), metallothioneins (MT1A, MT1F, MT1H), inhibitor, SLPI (P 0.05). IHC showed increased staining PSMD11 post-SCI, along peripheral region cells. also Atrogin-1 at membrane region. Thus, there profound increase components, metallothioneins, SLPI, within SCI. Increased levels specifically cell periphery, indicate that proteins this may be early targets degradation
