摘要: Systemic and localized ischemia reperfusion injury remain clinically relevant issues after organ transplantation contribute to dysfunctions, among which acute kidney is one of the most common. An in vitro test-circuit for normothermic perfusion porcine kidneys warm was used investigate antioxidant properties vitamin C during reperfusion. Vitamin known enhance microcirculation, reduce endothelial permeability, prevent apoptosis, inflammatory reactions. Based on current evidence about pleiotropic effects C, we hypothesize that might provide organ-protection improve graft function this model Methods: 10 from 5 Landrace pigs were perfused 6 h. For each experiment, both animal simultaneously with a 1:1 mixture autologous blood modified Ringer’s solution at 38 °C 75 mmHg continuous pressure. One treated 500 mg bolus injection into perfusate, followed by infusion 60 mg/h C. In control test circuit, an equal volume administered as placebo. Perfusate samples withdrawn distinct points time h gas analyses well measurement serum chemistry, oxidative stress capacity. Hemodynamic parameters urine excretion monitored continuously. Histological analyzed detect tubular- glomerular-injury. Results: administration perfusate significantly reduced (49.8 ± 16.2 vs. 118.6 23.1 mV; p = 0.002) perfusion, increased capacity, leading red cell protection hemoglobin concentrations (5.1 0.2 3.9 0.6 g/dL; 0.02) contrast placebo treatment. Kidney not different between groups (creatinine clearance vit C: 2.5 2.1 placebo: 0.5 mL/min/100 g; 0.9). Hypernatremia (187.8 4.7 176.4 5.7 mmol/L; 0.03), lower, but significant decreased fractional sodium (7.9 2 27.7 15.3%; 0.2) observed group. analysis did show differences glomerular groups. Conclusion: treatment capacity grafts, stress, preserved cells oxygen carrier like or attenuate damage. Nevertheless, due its antioxidative be beneficial supplement clinical protocols.
