摘要: Copper is an integral component of various enzymes, necessary for mitochondrial respiration and other biological functions. Excess copper related with neurodegenerative diseases as Alzheimer able to modify cellular redox environment, influencing its functions, signaling, catabolic pathways. Tryptophan degradation through kynurenine pathway produces some metabolites properties 3-hydroxykynurenine (3-HK) 3-hydroxyanthranilic acid (3-HANA). The imbalance in their production neuropathologies, where the common factors are oxidative stress, inflammation, cell death. This study evaluated effect these kynurenines on toxicity astrocyte cultures. It assessed CuSO4 effect, alone combination 3-HK or 3-HANA MTT reduction, ROS production, membrane potential (MMP), GHS levels, viability primary cultured astrocytes. Also, chelating was evaluated. results showed that decreased MMP, GSH levels while death increasing. Coincubation enhances toxic all markers tested except which abolished by kynurenines. Data suggest increased independent manner production.
