A novel approach to clonality analysis. Alterations in genomic methylation of Epstein-Barr virus transformed lymphocytes.

摘要: Summary We have studied eight different Epstein-Barr virus transformed cell lines (EBV-LCL) with respect to genomic methylation at a single locus, M-bcr (the major breakpoint cluster region of chronic myelogenous leukemia). Restriction digests the sensitive enzyme, Hpall, illustrated marked differences in patterns between various lines. Some displayed prominent allelic heterogeneity: within each these samples there were numerous M-bcr/Hpall fragments on Southern analysis. Other predominance: limited number predominated. Analysis Immunoglobulin JH rearrangements demonstrated close correlation clonal complexity (i.e. rearranged fragments) and heterogeneity fragments). conclude that analysis locus may offer novel approach clonality determination EBV lymphocytes.
