摘要: Restriction-modification system is present in bacteria to protect the cells against phage infection. Interestingly, bacteriophage MB78, a virulent of Salmonella typhimurium possesses restriction-modification system. Permissive host transformed with plasmid having genomic fragment MB78 carrying putative genes severely restrict growth 9NA. Growth also restricted some extent. However, temperate P22 not at all. Cloning has been done both orientations different vectors. The clones same orientation as that lacZ pUC19 are mostly unstable. those stable when cloned opposite orientation. Viability transformants strain-, orientation-, and medium-dependent. two have individually/separately. Hosts only modification gene do phages while hosts restriction do. former produces latter very unstable which were viable upto 36 h or so. colonies normal looking tiny, flat, looked distressed resembling much bacterial Amplification subsequent cloning expression vector will be carried out for characterization enzymes.
