摘要: The polyhydroxyalkanoic acid (PHA) biosynthetic gene locus was cloned and characterized from an Acinetobacter sp. isolated activated sludge. Nucleotide sequence analysis identified three clustered genes, phaAAc (encoding a beta-ketothiolase), phaBAc acetoacetyl coenzyme A reductase), phaCAc PHA synthase). In addition, open reading frame (ORF1) with potential to encode 13-kDa protein within this locus. of the putative translational product ORF1 does not show significant similarity any sequences in database. plasmid containing pha conferred ability accumulate poly-beta-hydroxybutyrate on its Escherichia coli host. These genes appear lie operon transcribed by two promoters upstream phaBAc, apparent constitutive promoter, second promoter induced phosphate starvation under pho regulon control. as well number additional transcription start points were combination primer extension promoter-chloramphenicol acetyltransferase fusion studies carried out or E. transformants.
