摘要: Objective: To investigate functional neural pathway organization and changes over-time in patients with Parkinson’s disease (PD) using advanced network-based techniques. Background: Graph theoretical analyses network science are powerful tools to study of the brain. Design/Methods: 146 PD performed clinical cognitive evaluations resting-state MRI at baseline every year for 4 years. Hierarchical cluster analysis identified two subtypes: 86 “early” 60 “mild-to-severe” patients. Within subtype, groups were identified: “early-motor-predominant” “early-diffuse”, latter having greater deficits more frequent non-motor manifestations. age- sex-matched controls assessments. connectomics assessed global local topological properties regional connectivity (FC) over-time. Results: “Early” showed a relatively preserved architecture At baseline, altered sensorimotor parietal areas relative controls. Longitudinal progressive deterioration features patients, specific involvement sensorimotor, frontal, temporal regions. widespread pattern decreased FC involving basal ganglia, networks was found “early-diffuse” cases. When compared between subtypes, five distinct patterns progression 1) different trend change subtypes (increase vs decrease); 2 & 3) similar or decrease), without difference groups; 4) but stable values over time subtypes; 5) no groups. Conclusions: might represent approach understand pathophysiological process associated progression.
