The central serotonin2B receptor as a new pharmacological target for the treatment of dopamine-related neuropsychiatric disorders: Rationale and current status of research

摘要: Abstract The serotonin2B receptor (5-HT2BR), which was first cloned and characterized in the rat stomach fundus, is most recent addition to 5-HT2R family. While its involvement regulation of gastrointestinal, vascular, pulmonary cardiac physiology has been widely investigated, functional role within central nervous system (CNS) received much less attention. Nevertheless, when considering data available literature with regards regulatory control exerted by 5-HT2BR on dopamine (DA) serotonin (5-HT) neuron activity, a very interesting picture emerges highlights key these receptors for future therapeutic strategies DA-related neuropsychiatric disorders. Thus, present review, compiling molecular, biochemical, electrophysiological behavioral findings from past twenty years, aims at providing sound analysis current knowledge supporting interest avenues. First, we recall neuroanatomical relevance 5-HT/DA interaction CNS. Thereafter, after short overview expression molecular properties 5-HT2BR, as well agonists antagonists market, will focus this 5-HT, DA neuroglia activity rodent brain. Finally, potential improved treatment schizophrenia drug addiction be discussed.
