摘要: Math1, a basic helix-loop-helix transcription factor homolog of the Drosophila atonal gene, is considered to be key for induction sensory hair cells (HCs) during development organ Corti or cochlea. Although embryonic stem (ES) are able produce HC-like cells, role Math1 in those has not been thoroughly elucidated. In present study, we introduced into ES order achieve efficient generation cells. carrying Tet-inducible Math1-ES were generated using Tet-On gene expression system. Embryoid bodies (EBs) formed absence doxycycline (Dox) 4 days allowed grow an additional 14 dishes presence 400 μg/ml Dox. At end 14-day cultures, approximately 10% EB outgrowths expressed HC-related markers myosin6, myosin7a, calretinin, α9AchR, and Brn3c (also known as Pou4f3) showed formation stereocilia-like structures, whereas few grown without Dox markers. Reporter assays Brn3c-promoter plasmid demonstrated positive regulation by Math1. Furthermore, such marker-positive derived from found incorporated developing inner ear after transplantation chick embryos. source ES-derived may important differentiating
