摘要: DNA vaccines are circular, double-stranded plasmid (pDNA) molecules, which capable of initiating the expression protein antigens interest when introduced into cells. For this purpose, pDNA contains an eukaryotic cassette consisting a transcriptional promoter, coding sequence derived from target antigen gene, and terminator (Fig. 73.1). Although many different promoters have been investigated, none shown to be clearly superior constitutive CMV IE promoter. can consist single genes on one plasmid, multiple plasmids, or combination above. In biscistronic tricistronic constructs, internal ribosomal entry sites (IRES) equivalent sequences, dual triple promoters, cleavable linkage regions in fusion proteins used for genes. Upon transfer cells, enters nucleus transcribes messenger RNA (mRNA) encoding interest. The identical wild-type pathogen, genetically modified improve immunogenicity and/or reduce toxicity host. may also contain antibiotic resistance gene bacterial origin replication growth propagation E. coli. Constructs using selection elements other than utilized.
