Calcitonin/calcitonin gene-related peptide transcription unit: tissue-specific expression involves selective use of alternative polyadenylation sites.

摘要: Different 3' coding exons in the rat calcitonin gene are used to generate distinct mRNAs encoding either hormone thyroidal C-cells or a new neuropeptide referred as gene-related peptide neuronal tissue, indicating RNA processing regulation is one strategy tissue-specific of expression brain. Although two use same transcriptional initiation site and have identical 5' terminal sequences, their termini distinct. The polyadenylation sites for located at end 4 6, respectively. Termination transcription after exon does not dictate production mRNA, because proceeds through both irrespective which mRNA ultimately produced. In isolated nuclei, appear be utilized; however, proximal (calcitonin) preferentially nuclei from tissues producing mRNA. These data suggest that mechanism dictating each involves selective alternative sites.
