Analysis of cocaethylene, benzoylecgonine and cocaine in human urine by high-performance thin-layer chromatography with ultraviolet detection: a comparison with high-performance liquid chromatography

摘要: Abstract Cocaine and ethanol are frequently used at the same time, resulting in formation of cocaethylene by transesterification. We studied capability high-performance thin-layer chromatography (HPTLC) to simultaneously detect cocaethylene, cocaine benzoylecgonine 16 urine specimens drug addicts, previously tested as positive for immunoenzymatic screening. Accuracy precision, well detection quantitation limits method, were evaluated comparison with liquid (HPLC). HPTLC limit was 1.0 μg/ml three compounds, whereas HPLC 0.2 cocaine, 0.1 cocaethylene. The relative standard deviation (RSD) ranged from 1.03 12.60% 1.56 16.6% intra- inter-day analysis, respectively. In case RSD intra-day precision 0.79 5.05%, it 1.19 10.64% precision. HPLC, is less expensive faster, requiring 2–3 h analyze 10–12 samples on a single plate. conclusion, suitable determinations analytes only high concentrations.
