摘要: Publisher Summary This chapter focuses on prolyl oligopeptidases. enzyme degrades a variety of proline-containing peptides by cleaving the peptide bond at carboxy side proline residues. Prolyl oligopeptidase is cytosolic serine peptidase that may be involved in metabolism hormones and neuropeptides. Hydrolysis proteases proceeds through formation an acyl-enzyme intermediate. The activity can measured spectrofluorometrically with Z-Gly-Pro-Nap (Z, benzyloxycarbonyl; Nap, 2-naphthylamine) substrate. displays strong dependence ionic strength reaction mixture. increase second-order rate constant approximately linear up to 0.3–0.5 M NaCl levels off higher salt concentrations. depends pH nature remarkably sensitive organic solvents, which shows even 1% acetonitrile inhibits about 50%. preferentially cleaves contains residue P1 position.
