Acquisition of a functional T cell receptor during T lymphocyte development is enforced by HEB and E2A transcription factors.

摘要: Summary The T cell receptor (TCR) is required for positive selection and the subsequent transition from CD4 + CD8 double-positive (DP) to or single-positive (SP) stage of αβ development. molecular mechanism that maintains DP fate prior acquisition a functional TCR not clear. We have shown here structurally functionally related transcription factors HEB E2A work together maintain control SP transition. Simultaneous deletion in thymocytes was sufficient independent TCR. Loss allowed cells bypass requirement TCR-mediated selection, downregulate DP-associated genes, upregulate SP-specific genes. These results identify as gatekeepers at development until αβTCR produced.
