The formin FMNL3 assembles plasma membrane protrusions that participate in cell–cell adhesion

摘要: FMNL3 is a vertebrate-specific formin protein previously shown to play role in angiogenesis and cell migration. Here we define the cellular localization of endogenous FMNL3, dynamics GFP-tagged during migration, effects suppression mammalian culture cells. The majority localizes punctate pattern, with >95% these puncta being indistinguishable from plasma membrane by fluorescence microscopy. A small number dynamic cytoplasmic patches also exist, which enrich near cell–cell contact sites fuse at sites. These appear be part larger membranes endocytic origin. On membrane, enriches particularly filopodia ruffles nascent adhesions. FMNL3-containing occur both cell–substratum interface contacts, latter 10-fold more stable. siRNA has two major effects: decrease compromised adhesion cells migrating as sheet. Overall our results suggest that functions assembly actin-based protrusions are specialized for adhesion.
