Mercury induced antinuclear antibodies in mice: characterization and correlation with renal immune complex deposits.

摘要: Female SJL and Balb/c mice were given subcutaneous injections of 1.6 mg HgCl2/kg body weight every third day for 2, 4, 8, or 12 weeks. Indirect immunofluorescence using HEp-2 cells as substrate showed that developed antinuclear antibodies (ANA) with a predominantly nucleolar weaker, homogeneous nuclear pattern after 4 weeks treatment. The antigen was sensitive to treatment trypsin RNAse, but the antibody not absorbed by calf liver RNA. responsible trypsin, DNAse, acid solution, reconstitution histones on treated did restore fluorescence. corresponding double-stranded single-stranded DNA, Crithidia luciliae assay negative. This suggests ANA is non-histone chromatin protein. No autoantibodies found in mice. Electron dense immune deposits containing IgG C3 mesangial-vascular mercury Acid eluate from kidneys contained tissue-bound strictly anti-nucleolar pattern, showing such make up part renal deposits. findings demonstrate induces polyclonal autoantibody response mice, restricted unknown specificity both cases leading complex kidneys.
