Intestinal and renal Na+/glucose cotransporters share common structures.

摘要: Polyclonal antibodies were raised to peptides selected from three different regions of the cloned rabbit intestinal Na+/glucose cotransporter. Western blot analysis was used identify fully mature protein in and renal brush borders. Two specifically identified a approximately 70-kDa band borders but did not immunoreact with membranes that do have Na(+)-dependent glucose transport activity. The immunoreactive proteins had an apparent isoelectric point between pH 4.7 6.8. also recognized similar-sized human seven other mammalian Similar bands four brush-border membranes, indicating shared epitopes cotransport proteins. In some species, e.g., lamb pig, epitope for one antibody missing both borders, suggesting this is essential function. These results suggest 1) cotransporter earlier biochemical studies, 2) there close structural similarity cotransporters, 3) structure these has been conserved during evolution.
