摘要: The target specificity of DNA strand transfer mediated by human immunodeficiency virus type 1 integrase was examined in vitro with synthetic oligonucleotides. Although insertion occurred at most locations the target, some sites were preferred over others least 15-fold. Changing nucleotide sequence changed distribution complex ways, which included changes preference distant from alteration. Alignment sequences revealed that adenosine is adjacent to site. Strand within 2 nucleotides 3' end and 3 5' target. This suggests only or flanking site are required for integration; such restricted contact would allow insert two ends viral into closely spaced host DNA, consistent concerted vivo integration reaction generates a 5-bp duplication.
