摘要: Raman spectroscopy can be a powerful tool for the characterization of modified amino acids and proteins. In addition to potential quantitative results, it offers advantage not requiring any sample preparation. Modification amines thiols on proteins are common reactions used medical, biological, food, agricultural purposes. We hypothesized that spectrum could quantify would more informative than typical techniques such as ninhydrin test. To prove hypothesis, alanine, cysteine, lysine were with ethyl vinyl sulfone (EVS) using nucleophilic reaction known Michael product was characterized spectroscopy. The results compared ultraviolet-visible based analysis acids. able discern site-specific suggested acid moieties substituted predicted test alone. Substitution full protein ovalbumin EVS showed similar results. substitution primary but detect secondary remaining after loss amine. Copyright © 2010 John Wiley & Sons, Ltd.
