摘要: The LNCaP human prostate cancer cell line is dependent on androgen for in vitro growth. To discover genes that may be responsible progression of from hormone dependence to independence, we transfected cells with expression vectors contained either the v-rasH or c-rasH gene under control cadmium (Cd2+)-inducible metallothionein-IIA promoter. Numerous derivative lines were isolated which manifested inducible rasH p21 protein when treated Cd2+. None found have an altered growth phenotype. Several expressing hormone-independent culture 10(-7) M Cd2+ . induction was also shown increase anchorage-independent colony formation v-rasH-expressing tested. Expression a dominant mutated oncogene can change hormone-dependent phenotype cells.
