Critical role of GSH in Sulfur Mustard-induced Oxidative Stress and Cytotoxicity in Human Skin Fibroblast Cell Line

摘要: In this study the role of glutathione (GSH) in sulfur mustard -induced oxidative stress and cytotoxicity, human skin fibroblast cell line (HF2FF) was evaluated. Sulfur mustard-induced superoxide radical hydrogen peroxide formation were evaluated by determination dismutase catalase activity lysate. The cytotoxicity estimated lactate dehydrogenase leakage. intracellular GSH content modulated N-acetylcysteine (NAC), a precursor, buthionine sulfoximine (BSO), specific synthesis inhibitor. It found that exposure led to dose-and time-dependent decrease HF2FF cells. NAC increased level protected cells against reactive oxygen species contrast, pretreatment depleted cellular enhanced susceptibility cytotoxic effects mustard. These results indicated plays critical protecting mustar-induced injury, most probably through its antioxidant activity.
