Arabinonucleic Acids: 2′-Stereoisomeric Modulators of siRNA Activity

摘要: We have investigated, for the first time, short interfering duplexes containing arabinonucleotides (ANA; 2′-stereoisomer of RNA), as well combinations ANA with RNA, and their 2′-fluorinated derivatives 2F-ANA and/or 2′F-RNA. The results show that is especially accommodated in sense strand small RNA (siRNA) duplexes, which can be extensively modified little effect on potency. Furthermore, combining 2′F-ANA siRNA passenger strands, particularly patterns bias duplex thermal stability, produces similar (and sometimes enhanced) potency compared native siRNA. Effective modification were identified against firefly luciferase screens HeLa cells then applied to knockdown down-regulated renal cell carcinoma (DRR), a novel clinically tractable target treatment glioblastoma.
