Quantitative Distribution Studies in Animals: Cross-Validation of Radioluminography versus Liquid-Scintillation Measurement

摘要: The results of a cross-validation the radioluminography (RLG) and liquid scintillation counting (LSC) methods are presented. for determination radioactivity concentrations were compared in 16 organs, after administration (14)C-labeled substances to rats. LSC measurements two kinds used as reference RLG: (1) quantitative conventional dissection (interindividual comparison) (2) tissue punches taken from whole-body sections they had undergone RLG measurement (intraindividual comparison). Blood standards containing known calibration. For statistical evaluation log-linear regression analysis paired concentration values organ-specific 95% confidence intervals log-transformed RLG/LSC quotients compared. most slopes lines means within defined equivalence range 0.80-1.25. Deviations distinctly smaller intraindividual comparison. some however, it became clear that found affected by self-absorption differences sample preparation (LSC). In conclusion, quantification with is reliable reproducible method comparable precision greater accuracy respect localization, (dissection).
