摘要: Cytokines are low molecular weight mediators of the immune system. Because their properties and biological roles, they have been used as diagnostic reagents for human animal diseases, well prognostic tests prophylactic treatments. Concerning this kind treatments, there is uncertainty to how cytokines effective after oral administration. They probably act on lymphoid tissues (palatine pharyngeal tonsils) trigger a cascade events leading activation system control inflammatory in organs. Owing above, aim our project was develop reliable vitro model related crucial interactions between cells, terms homeostatic regulation response antibody production. In project, we focused attention IFN-α The study divided into 4 steps: 1) Isolation culture pig tonsil lymphocytes; 2) evaluation IPEC-J2 cells reporter anti-inflammatory actions interferon-alpha; 3) A cell evaluating oral, low-dose treatments; 4) Disease-dependent modulation phenotypes. first steps procedures isolation which were validated use functional immunoassays. Our results indicate that can be employed within 2 months from freezing maintain suitable conditions recovery, vitality release vitro. Tonsil mononuclear also showed ability secrete antimicrobial peptides respond immunological stimuli. second part (a continuous line porcine intestinal epithelial cells) IFN-α. Three different experimental (oxidative stress, response, amplification signals) selected evaluate effects recombinant IFN-α1 (rIFN-α) natural preparations (nIFN-α). IFNs under significantly cells. particular, rIFN-α shown down-regulate interleukin (IL)-8, IL-1β, tumor necrosis factor (TNF)-α, β-defensin 1 genes either directly, or indirectly through messengers released by IFN-α-treated With regard IL-6, only could regulate expression cytokine. useful tool investigating regulatory type I thereof. third step developed an interaction types IFNs-α at / moderate concentrations with respect three fundamental actions: IgA (up down-regulation,…
