摘要: qPCR has become increasingly popular for microbial water quality testing because it is faster, more specific, and flexible than culture-based methods. However, method limitations such as quantification bias introduced by reliance on standards susceptibility to PCR inhibitors are major obstacles implementation in testing. This requires accurate of rare targets because environmental waters often contain inhibitors. Digital offers the opportunity maintain qPCR's advantages over methods while ameliorating two limitations: necessity run standard curves high inhibition. Here we describe a complete simultaneous general indicator (Enterococcus spp.) human-associated fecal marker environmental waters. The includes sampling filtration capture bacteria, DNA extraction from bacteria captured filter, droplet digital quantify genetic markers indicative contamination.
