Effect of Insulin and Glucagon on Hepatocellular Alanine Uptake in Normal and Streptozocin-Induced Diabetic Rats

摘要: Hepatocytes from normal and streptozocin-treated diabetic rats were dispersed by collagenase perfusion. Cells suspended in Ca-Mg free phosphate buffer containing oxamic acid treated with insulin, glucagon and/or cycloheximide prior to measurement of 14C-alanine transport. Samples withdrawn at specified intervals, centrifuged, hepatocellular uptake was monitored alanine intracellularly extracellularly. Alanine insulin-treated cells increased (86%) compared controls. However, cycloheximide-treated cells, decreased 25 %. Cycloheximide, the presence blocked induced insulin treatment. Diabetic showed a % increase cells. Insulin treatment enhanced (67%), while it 13 No change observed when simultaneously. enhances transport hepatocytes contrary effect cycloheximide. data disagrees “down-regulation” concept. The exhibited may be explained an plasma levels which has been shown potent stimulator carrier receptors.
