摘要: The thalamus is a diencephalic structure that plays crucial roles in relaying and modulating sensory motor information to the neocortex. develops dorsal part of neural tube at level caudal forebrain. However, molecular mechanisms are essential for thalamic differentiation still unknown. Here, we have succeeded generating neurons from mouse embryonic stem cells (mESCs) by modifying default method induces most-anterior type self-organizing culture. A low concentration caudalizing factor insulin MAPK/ERK kinase inhibitor enhanced expression forebrain markers Otx2 Pax6. BMP7 promoted an increase precursors such as Tcf7l2+/Gbx2+ Tcf7l2+/Olig3+ cells. mESC began express glutamate transporter vGlut2 axon-specific marker VGF, similar mature projection neurons. extended their axons cortical layers both organotypic culture subcortical transplantation.
