Capsaicin-induced apoptosis of glioma cells is mediated by TRPV1 vanilloid receptor and requires p38 MAPK activation

摘要: We provide evidence on the expression of transient receptor potential vanilloid type-1 (TRPV1) by glioma cells, and its involvement in capsaicin (CPS)-induced apoptosis. TRPV1 mRNA was identified quantitative RT-PCR U373, U87, FC1 FLS with U373 cells showing higher, lower as compared normal human astrocytes. By flow cytometry we found that a substantial portion both astrocytes, U87 express protein. Moreover, analyzed at protein levels tissues different grades. gene inversely correlated grading, marked loss majority grade IV glioblastoma multiforme. also described CPS trigger apoptosis but not cells. CPS-induced involved Ca(2+) influx, p38 extracellular signal-regulated mitogen-activated kinase activation, phosphatidylserine exposure, mitochondrial permeability transmembrane pore opening dissipation, caspase 3 activation oligonucleosomal DNA fragmentation. functionally implicated these events they were markedly inhibited antagonist, capsazepine.
