Assessment of endogenous reference gene suitability for serum exosomal microRNA expression analysis in liver carcinoma resection studies.

摘要: Abstract Serum exosomal microRNAs (miRNAs) have received considerable attention as potential biomarkers for tumor diagnosis. Reverse transcription‑quantitative polymerase chain reaction (RT‑qPCR) is commonly used to detect miRNA expression levels in various types of cancer. One prerequisite valid RT‑qPCR data the correct normalization miRNAs stably expressed endogenous reference genes (RGs). The study liver carcinoma resection requires use reliable RGs order assess serum target miRNAs. However, assessment RG suitability optimum analysis has yet be investigated. present investigated stability 10 candidate RGs. included eight (miR‑16, miR‑103, miR‑191, let‑7a, miR‑26a, miR‑221, miR‑181a, and miR‑451) two small RNAs (5S U6). values were calculated using following algorithms: geNorm, NormFinder, BestKeeper, comparative ΔCt method. overall ranking obtained from these analyses revealed that miR‑26a appropriate internal patients with hepatocellular carcinoma. In addition, miR‑221 let‑7a combined, recommended by or increased accuracy interpretation hepatopathy studies.
