摘要: The genome of Saccharomyces cerevisiae encodes 35 members a family proteins thattransport metabolites and substrates across the inner membranes mitochondria. They includethree isoforms ADP/ATP translocase phosphate citrate carriers. At startof our work, functions remaining 30 were unknown. We areattempting to identify these by overexpression in specially selectedhost strains Escherichia coli that allow carriers accumulate at high levels formof inclusion bodies. purified are then reconstituted into proteoliposomes wheretheir transport properties studied. Thus far, we have identified dicarboxylate,succinate-fumarate ornithine Bacterial functional identification, togetherwith characterization yeast knockout strains, has brought insight physiologicalsignificance transporters. dicarboxylate carrier sequence been used toidentify orthologous protein Caenorhabditis elegans and, turn, this latter sequencehas establish human ortholog.
