Intravitreal acetylsalicylic acid in silicone oil: pharmacokinetics and evaluation of its safety by ERG and histology.

摘要: Background: A new method of intravitreal drug delivery acetylsalicylic acid (AS) in silicone oil was investigated for safety and its pharmacokinetics the posterior pole eye. Methods: The AS mixed to a concentration 1.67 mg/ml. After vitrectomy, 15 NZW rabbits received an injection AS/silicone suspension. Clinical examination, pre- postoperative electroretinography (ERG) histology were performed. distribution salicylic determined by HPLC analysis at 6 h, 24 h 5 days optic nerve, retina, choroid, vitreous, blood. Results: examination revealed no adverse effects or signs toxicity. ERGs showed significant difference between results. concentrations demonstrated peak values residual vitreous (640.0 µg/ml), choroid (446.0 ng/mg) retina (281.3 h. At decreased 20.9 µg/ml 38.5 ng/mg retina. retinal level still 48.1 ng/mg. Conclusions: administration loaded is safe results high segment eye while maintaining low blood levels.
