Proteomic Analysis of Bovine Brain G Protein γ Subunit Processing Heterogeneity

摘要: We characterized the variable processing of G protein γ subunit isoforms associated with bovine brain proteins, a primary mediator cellular communication. Gγ subunits were isolated from purified proteins and by Edman sequencing, MALDI MS, chemical and/or enzymatic fragmentation assayed MS/MS sequencing. Multiple forms six different detected. Significant variation in was found at both amino termini particularly carboxyl proteins. All contain carboxyl-terminal CAAX motif for prenylation, proteolysis, carboxymethylation. Characterization these indicates significant variability normal all steps prenylation reaction, including new prenyl resulting cysteinylation terminus. These results have multiple implications intracellular signaling mechanisms role cell signaling, site action consequences drugs that target modification.
